Lisa Edelstein is one of the three children of Alvin and Bonnie Edelstein. Her father works as a pediatrician at the Chilton Memorial Hospital in New Jersey. Having been raised in Wayne, New Jersey, Lisa graduated from the Wayne Valley High School, in 1984. At the age of 18, Lisa moved to New York City where she joined studied theatre at the New York University's Tisch School of the Arts.

During the time she was living in New York, Lisa mixed up with the club scene, where she was known only as "Lisa E.". She created so much of a stir in the community that, writer and fellow celebutante James St. James gave her the name of New York City's "Queen of Downtown".

Lisa is a vegetarian and does not eat meat or wear things or accessories made from animal products. Being an animal lover, she spends time at the animal sanctuary named Best Friends. She is also a yoga enthusiast and devotedly practices Mysore style Ashtanga Yoga, six days a week. She takes pleasure in writing, composing music and painting. She also speaks Spanish. Lisa currently lives in Los Angeles and owns a cat and three rescue dogs named, Sandwich, Wolf-E and Bumpa.

Lisa started her career by writing, composing and starring in an original musical named Positive Me. It dealt with the issue of growing crisis of Aids, in the 1980’s. She then did a brief stint as the host on Awake on the Wild Side for MTV in 1990. This was followed by many guest appearances in various popular comedy shows on television. These included Mad About You, Wings, The Larry Sanders Show and Seinfeld.

Soon Lisa started getting bigger roles in TV dramas. She portrayed major characters on shows like Relativity, The West Wing, Ally McBeal, Felicity, House MD etc., while continuing her guest appearances on shows like ER, Frasier, Just Shoot Me!, Without a Trace and Judging Amy. In addition Lisa has also done voice over acting for shows like, King of the Hill, American Dad!, Superman: The Animated Series etc.

Lisa has also made a name for herself in the movie world where she has portrayed roles in movies like, L.A. Without a Map, 30 Days, What Women Want, Say Uncle, Grilled, My Lunch with Larry, as well as various other movies.

In 2005, Lisa won the Satellite Award under the category of Outstanding Actress in a Supporting Role in a Series, Mini-Series or Motion Picture Made for Television, for her role in House MD. In 2009, she was nominated for the Screen Actor Guild Award under the category of Outstanding Performance by an Ensemble in a Drama Series, along with her other co-stars for House MD.

In House MD, Lisa plays the role of Dr. Lisa Cuddy. She is the Dean of Medicine, and an endocrinologist. She is considered to be one of the friends of Dr. House and is one of the few characters who can not only match wits but also argue with the fast-talking Dr. House. She is also amongst the few who is able to tolerate Dr. House's rude manner, strange requests, and his several horrible habits. Even though, Lisa often disapproves of the methods used by Dr. House, she trusts that his decisions are taken keeping the best interest of his patients in mind. She often is left to shoulder the burden of Dr. House's disputed medical practices.
